Aberrant receptor expression or functioning of the epidermal growth factor receptor (Erbb) family plays a crucial part in the development and evolution of cancer. Inhibiting the signalling activity of individual receptors in this family has advanced the treatment of a range of human cancers. In this Review we re-evaluate the role of two important family members, ERBB2 (also known as HER2) and ERBB3 (also known as HER3), and explore the mechanisms of action and preclinical and clinical data for new therapies that target signalling through these pivotal receptors. These new therapies include tyrosine kinase inhibitors, antibody–chemotherapy conjugates, heat-shock protein inhibitors and antibodies that interfere with the formation of ERBB2–ERBB3 dimers.


We want the magic combo to be discovered.

Her-2+++; lumpectomies

Aug 06: Stage IV
liver mets: 6 tumours
July 06 to Jan 07: 2*FEC+6*Taxotere; 3*TACE; LITT
March 07- Sept 07: Vaccination trial (phase 2, peptide based) at the UW (Seattle).
Herceptin since 2006
NED til Oct 09
Recurrence Oct 2009: to internal mammary gland since October 2009 missed on Oct and March 2010 scan.. palpable nodes in May 2010 when I realised..
Nov 2011:7 mets to lungs progressing fast failed hercp/tykerb/xeloda combo..

superior vena cava blocked: stent but face remains puffy

April 2012: Teresa Trial, randomised to TDM1
Nov 2012 progressing on TDM1
Dec 2012 blockage of my airways by tumours, obliteration of these blocking tumours breathing better but hoping for more- at mo too many tumours to count in the lungs and nodes.

Dec 2012 Starting new trial S-222611 phase 1b dual egfr her2+ inhibitor.

Aug 2010: diagnosed stage 3b, 4 mo. after birth of son. 29 yrs old and breastfeeding, ER/PR-, Her-2+ started Neoadjuvant therapy: 4x FEC, 10x abraxane & Herceptin
Feb 2011: L mx with recon. Path. showed only DCIS but 4/10+ nodes.
March 2011: 6 wks rads.
Mother passed, lower back pain.
Late May 2011: Bone mets but organs clear; Tykerb, Xeloda, Xgeva. Stopped Herceptin. Implant infected: removed implant.
October 2011: Bone progression; Gemzar and Carboplatin & restarted Herceptin.
Jan 2012: Progression, re-classified as ER+; Tykerb, Herceptin, Zoladex & Femara. Anti-E is working!
May 2012: ovaries out, markers stable but elevated. Cont. Herceptin, Tykerb, Xgeva & Femara.
Dec 2012: aromasin
Jan 2013: faslodex, herceptin, tykerb
Jun: Kadcyla
Aug: Rads to hip, then Perjeta, Herceptin & Taxotere
Nov 2013: Perjeta, Herceptin, Halaven
Early 2014: Affinitor, Aromasin, Perjeta, Herceptin.
June 2014: Estradiol, Perjeta, Herceptin
Aug 14: Tamoxofin, H & P

With all the inhibitors/ma and various her2/3/4 available there is a magic combo somewhere that people could tolerate for a short time at least. Tdm1 and neratinib for one with alternative others like hsp90 need to be urgently tested. It seems ridiculous that everyone agrees the target need to be simultaneously blocked but combo trials are taking so so long..

Wish I slept well like your gorgeous puppy!
On a serious note thanks for posting.It is so frustrating to think that there may be a combo to treat us all but due to costs it's not available and the trials to assess it do not get done.If more negotiation globally was done with the drug companies about pricing maybe we would have more options.

How you got any news about treatment options yet? Would you be able to travel abroad for a trial or even to another centre in England?
Keeping my fingers x for you to get some positive news.

I know maybe that is where advocacy should start for her2 sisters and organise ourselves as you mentioned earlier like hiv+ group did to make the science move quicker: we definitely have a very strong case..


Reviewing and applying to trials at the moment. Get scan reviewed so they are assessing the rate of progressions..everything is taking so long.. but yes I can still travel anywhere.
