PARP inhibitor trial
Just a thought.
I'm borderline HER2+, and I often wonder whether that almost makes me triple-negative bc and whether I might benefit from TNBC therapies.
My lung met was HER2+, but my brain MET was HER2-.
PARP inhibitor trials for TNBC have been in the news lately showing phase II BSI-201 trial results and a new BSI-201 phase III trial, as well as a TNBC phase II trail results of a related PARP inhibitor drug, olaparib, for woman who are BRCA1 or BRCA2 positive.

I was told that I was borderline HER2+, and generally that means treating for HER2+, just to be on the safe side.

Diagnosed stage 2b in July 2003 (2.3 cm, HER2+, ER-/PR-, 7+ nodes). Treated with mastectomy (with immediate DIEP flap reconstruction), AC + T/Herceptin (off label). Cancer advanced to lung in Jan. 2007 (1 cm nodule). Started Herceptin every 3 weeks. Lung wedge resection April 2007. Cancer recurred in lung April 2008. RFA of lung in August 2008. 2nd annual brain MRI in Oct. 2008 discovered 2.6 cm cystic tumor in left frontal lobe. Craniotomy Oct. 2008 (ER-/PR-/HER2-) followed by targeted radiation (IMRT). Coughing up blood Feb. 2009. Thoractomy July 2009 to cut out fungal ball of common soil fungus (aspergillus) that grew in the RFA cavity (most likely inhaled while gardening). No cancer, only fungus. Removal of tiny melanoma from upper left arm, plus sentinel lymph node biopsy in Feb. 2016. Guardant Health liquid biopsy in Feb. 2016 showed mutations in 4 subtypes of TP53. Repeat of Guardant Health biopsy in Jana. 2021 showed 3 TP53 mutations, BRCA1 mutation and CHEK2 mutation. Invitae genetic testing showed negative for all of these. Living with MBC since 2007. Stopped Herceptin Hylecta (injection) treatment in March 2020. Recent 2023 annual CT of chest, abdomen and pelvis and annual brain MRI showed NED. Praying for NED forever!!

Thanks for offering the articles but I've read about triple negative cancer in a few places. Perhaps also in Cure, too. It's been in the news lately. I can probably check further into it on the Internet, and when I see my "other" oncologist soon, who specializes in bc, I'm going to ask him. I believe my HER2 status was very close to the cut-off point, or perhaps right on that point, as noted in the trial inclusion criteria.

I am trying to explore this also since my recurrence was triple negative. I am having my mastecomy tissue retested and will know more in a few weeks. I also need to get the BRAC testing done.
I know of a lady with her2 and BRAC who is doing well on this trial.

When I find out more from the onc I'll let you know.
TNBC is a bit confusing to me. I think it might be a "biological subset" of its own and therefore not apply at all (that is, either you have it or you don't). But sometimes therapies can work anyway. I intend to ask the onc.
