In this study, we unexpectedly observed exquisite susceptibility of HER2+ breast cancer cells to PARP inhibitors alone independent of an inherent HR deficiency. HER2 overexpres- sion itself was sufficient to render breast tumor cells suscep- tible to PARP inhibition. The mechanistic insight of this intriguing sensitivity is described in this report and involves attenuation of NF-kB signaling driven by HER2. Our results suggest that PARP inhibitors may be useful for sporadic HER2þ breast cancer patients.
