In this Review we re-evaluate the role of two important family members, ERBB2 (also known as HER2) and ERBB3 (also known as HER3), and explore the mechanisms of action and preclinical and clinical data for new therapies that target signalling through these pivotal receptors. These new therapies include tyrosine kinase inhibitors, antibody–chemotherapy conjugates, heat-shock protein inhibitors and antibodies that interfere with the formation of ERBB2–ERBB3 dimers. With all the inhibitors/ma and various her2/3/4 available there is a magic combo somewhere that people could tolerate for a short time at least. Tdm1 and neratinib for one with alternative others like hsp90 need to be urgently tested. It seems ridiculous that everyone agrees the target need to be simultaneously blocked but combo trials are taking so so long..

Wish I slept well like your gorgeous puppy! On a serious note thanks for posting.It is so frustrating to think that there may be a combo to treat us all but due to costs it's not available and the trials to assess it do not get done.If more negotiation globally was done with the drug companies about pricing maybe we would have more options. How you got any news about treatment options yet? Would you be able to travel abroad for a trial or even to another centre in England? Keeping my fingers x for you to get some positive news. Ellie |

I know maybe that is where advocacy should start for her2 sisters and organise ourselves as you mentioned earlier like hiv+ group did to make the science move quicker: we definitely have a very strong case..
Reviewing and applying to trials at the moment. Get scan reviewed so they are assessing the rate of progressions..everything is taking so long.. but yes I can still travel anywhere. |
