OPEN ACCESS: EDITORIAL: Whither HER2-Related Therapeutics?
[Journal of Clinical Oncology]
It is apparent that increasing knowledge of molecular signaling pathways regarding HER2/Neu amplified breast cancer biology has paved the way toward novel, innovative targeted combination therapeutics. The multiplicity of potential targets along the PI3K-AKT-mTOR or HER2-GRB7-RAC1 pathway requires careful preclinical evaluation to ensure optimal activity, minimal toxicity, and lack of resistance. The sheer multiplicity of approaches outlined above will need to correspond to measurements of driving molecular signaling pathways on an individual patient basis. In the meantime, the patient faces a panoply of HER2-directed therapies and state-of-the-art clinical trials, yielding better responses, survival, quality of life, and real hope of a cure.

12-7-09 Navelbine added to Herceptin/Zometa.
3-23-10 Ten days of rads to RT femur. Completed.
4-05-10 Quit Navelbine--Herceptin/Zometa alone.
5-4-10 Appt. with Dr. Slamon to see what is next? Waiting on FISH results from femur biopsy.
Results to FISH was unsuccessful--this happens less then 2% of the time.
7-7-10 Recurrence to RT axilla again. Back to UCLA for options.

Yeah!! I remember reading that an American breast onc professor last year said that he believed in 5 years no women would need to die from her 2 bc. There may not be a cure but that they would have the ability to control it.
Praying that 5 years becomes this year.

Yes, there is great promise for curing this disease. We all are praying that it comes sooner than later. On the other hand, the "panoply of HER2-directed therapies" now faced by the patient is almost too much to bear at times as we try to sort them all out. Those of us who have oncologists and groups like this one that can intelligently help guide us with the choices are the fortunate ones.

I have been following the Herceptin/PI3 kinase inhibitor combination for a while. Check out this poster from a company named ARRAY which makes a PI3 kinase inhibitor called ARRY 380. It shows that, in lab mice, this combo had a 100% cure rate!

ARRY 380 is currently in clinical trial as a single agent at M.D. Anderson.

However, for those of you interested in a clinical trial combining BOTH a version of Herceptin (T-DM1) and a PI3 kinase inhibitor, Dana Farber currently has one in a Phase I trial. I met with Dr. Ian Krop in December to discuss joining but he suggested I wait until it reaches Phase II as it is coming to UCSF - where I live. Here is a link to the Dana Farber trial:

Had dose dense neoadjuvant AC,Taxol then mastectomy,radiation+xeloda+Herceptin.

My onc has been going on about P13 inhibition each time I meet him. He is convinced that harnessing this would result in a quantum shift in treatment!!
So girls i am saving up my money so I can come and see you all if a trial becomes available!! Might even manage a chocolate with Laurel.

Combo with Honokiol HERE

IP-6 HERE

I have seen a form of IP-6 in Walgreens.

Seems like liposomal versions of supplements are preferrable when available.

Laurel - I will be there to celebrate the cure with you - chocolate martini in hand! It is coming, I KNOW IT!

Julie - I asked Melisko when the trial was coming to UC and she said "a few months" so that could mean 2 or 6, I don't really know. She was going to ask Rugo. I'd ask them about it the next time you are there. Let me know what they say. I don't actually go to UC for treatment but I occasionally consult with Melisko for second opinions.

IP - 6 - I used to get a good one at my local GNC store. I see that there are a few different trustworthy companies offering it at my local supplements specialty store. I don't take it now.

Personally, I do not get any supplements at Walgreens. Too many formulations from who knows where. I will pay a little more for best quality after what I have been through!

Looks like some exciting trials not far down the road. Please, girls, keep us posted.

MY STORY SO FAR ~~~~
Found suspicious lump 9/2000
Lumpectomy, then node dissection and port placement
Stage IIB, 8 pos nodes of 18, Grade 3, ER & PR -
Adriamycin 12 weekly, taxotere 4 rounds
36 rads - very little burning
3 mos after rads liver full of tumors, Stage IV Jan 2002, one spot on sternum
Weekly Taxol, Navelbine, Herceptin for 27 rounds to NED!
2003 & 2004 no active disease - 3 weekly Herceptin + Zometa
Jan 2005 two mets to brain - Gamma Knife on Jan 18
All clear until treated cerebellum spot showing activity on Jan 2006 brain MRI & brain PET
Brain surgery on Feb 9, 2006 - no cancer, 100% radiation necrosis - tumor was still dying
Continue as NED while on Herceptin & quarterly Zometa
Fall-2006 - off Zometa - watching one small brain spot (scar?)
2007 - spot/scar in brain stable - finished anticoagulation therapy for clot along my port-a-catheter - 3 angioplasties to unblock vena cava
2008 - Brain and body still NED! Port removed and scans in Dec.
Dec 2008 - stop Herceptin - Vaccine Trial at U of W begun in Oct. of 2011
STILL NED everywhere in Feb 2014 - on wing & prayer
7/14 - Started twice yearly Zometa for my bones
Jan. 2015 checkup still shows NED
2015 Neuropathy in feet - otherwise all OK - still NED.
Same news for 2016 and all of 2017.
Nov of 2017 - had small skin cancer removed from my face. Will have Zometa end of Jan. 2018.
