Chloroquine - Sounds promising!
Hi All - I recently read this article in Cure magazine talking about the common anti-malarial drug, Chloroquine, as a potential destroyer of cancer stem cells. It all sounds very promising. There will soon be a multicenter clinical trail using it in triple negative bc:

Intrigued by the article, I did some further research and found some promising preliminary studies such as this one, discussing Chlroquine as a HER2 disruptor:

So should I plan a trip to Africa so I can get my Chloroquine prescription now? Why not? Its already FDA approved. I've got half a mind to just try this out myself. Does anyone have any experience with this or thoughts? I took this drug years ago when traveling through Central America and other than a few lucid dreams, it really didn't seem to have any negative side effects.

Looks like anti malaria treatment generally have action on cancer cell since artesunate/ artemisinim is also found to be anti cancerous.
these a thread on it but not sure how to post it
Also it seems that a lot of things that are anti HIV and also anti cancerous- Chloroquine is also on trial against HIV

I took plaquenil (a form of this drug) for lupus many years ago. The only side effect I can remember is that it can cause problems with your eyes. I had to see an opthalmologist every three months as long as I took it.

5/21/10:port installed,TCH chemo (6 x 3 wks); Herc,-1yr; 33 rad tx after chemo
07/2010: port not working-2nd port didnt'work;3rd port opposite side.
07/2010: 2 weeks after 3rd port surgery, threw 3 pulm. emboli-IVC filter installed; warfarin
08/2010: hospitalizations w/3 of chemos; decision to stop after 4th-on to radiation in Oct 2010;Herc cont.
12/03/2010 - finished 33 rads Hooray!! cont. Herc. every 3 wks
4/2011 - pneumonia ??? Nope-radiation pneumonitis. No more Herc.
5/2011 - NED!!! port out.
8/2011 - clean PET & CT scans.Still NED
7/2012 - Still NED/very blessed.
2/2013 - 6 mos checkup-all clear. CA2729 down frm 13 to 11.

Courtney, I would be all for trying out a new drug except that there is the potential that it would interfere with your current therapy. What are you taking right now? I'd run it by some professional first, either your oncologist or chemo pharmacist. Sometimes things as seemingly harmless as grapefruit can decrease the efficacy of our life-saving chemo drugs.
I also took the drug when I was traveling in South America, and like you don't remember any side effects from it.

Oh and one other thing- you should totally call, email or write a letter to any of the authors or doctors in charge of the study and ask their opinion. Most researchers will respond and offer good advice, as it is in their interest to validate their studies with additional patient experiences.

This is a very interesting thread for me. I also took the drug Plaquinil/hydroxycloroquin for a few years to treat "unspecified inflammatory syndrome." This began not long after I finished my treatment for liver mets. First I was taking Celebrex as a COX2 inhibitor, prescribed by my treating med oncologist.
Both these drugs now have some research for anti-HER2. I do not know how close the formulation for hydroxycloroquin is to the cloroquine mentioned in the article, but it would be interesting if they are pretty close.
Maybe some of our Research Assistants can find a molecular comparison.

MY STORY SO FAR ~~~~
Found suspicious lump 9/2000
Lumpectomy, then node dissection and port placement
Stage IIB, 8 pos nodes of 18, Grade 3, ER & PR -
Adriamycin 12 weekly, taxotere 4 rounds
36 rads - very little burning
3 mos after rads liver full of tumors, Stage IV Jan 2002, one spot on sternum
Weekly Taxol, Navelbine, Herceptin for 27 rounds to NED!
2003 & 2004 no active disease - 3 weekly Herceptin + Zometa
Jan 2005 two mets to brain - Gamma Knife on Jan 18
All clear until treated cerebellum spot showing activity on Jan 2006 brain MRI & brain PET
Brain surgery on Feb 9, 2006 - no cancer, 100% radiation necrosis - tumor was still dying
Continue as NED while on Herceptin & quarterly Zometa
Fall-2006 - off Zometa - watching one small brain spot (scar?)
2007 - spot/scar in brain stable - finished anticoagulation therapy for clot along my port-a-catheter - 3 angioplasties to unblock vena cava
2008 - Brain and body still NED! Port removed and scans in Dec.
Dec 2008 - stop Herceptin - Vaccine Trial at U of W begun in Oct. of 2011
STILL NED everywhere in Feb 2014 - on wing & prayer
7/14 - Started twice yearly Zometa for my bones
Jan. 2015 checkup still shows NED
2015 Neuropathy in feet - otherwise all OK - still NED.
Same news for 2016 and all of 2017.
Nov of 2017 - had small skin cancer removed from my face. Will have Zometa end of Jan. 2018.

Dec'08~ Brain MRI- 4 spots reduced to punctate size, large spot shrunk by 3mm, CT of torso clear/pelvis spot stable
June'09~ new 3-4mm left cerrebellar spot zapped with IMRT targeted rads
Sept'09~ new 6mm & 1 cm spots in pituitary/optic chiasm area. Rx= 25 days of 3D conformal fractionated targeted IMRT to the tumors.
Oct'09~ 25 days of low dose 3D conformal fractionated targeted IMRT to the bone mets spot on rt. iliac crest that have been watching for 2 years. Added daily Aromasin back into treatment regimen.
Apr'10~ Brain MRI clear! But, see new small spot on adrenal gland. Change from Aromasin back to Tamoxifen.
June'10~ Tumor markers (CA15.3) dropped from 37 to 23 after one month on Tamoxifen. Continue to monitor adrenal gland spot. Remain on Tykerb/Herceptin/Tamoxifen.
Nov'10~ Radiate positive mediastinal node that was pressing on recurrent laryngeal nerve, causing paralyzed larynx and a funny voice.
Jan'11~ MRI shows possible activity or perhaps just scar tissue/necrotic increase on 3 previously treated brain spots and a pituitary spot. 5 days of IMRT on 4 spots.
Feb'11~ Enrolled in T-DM1 EAP in Denver, first treatment March 25, 2011.

Wow, that is really interesting, Steph. I wonder if that had anything to do with your success? I started the Chloroquine yesterday and hope that it is helping to get rid of this mess. Will keep y'all posted, as usual.

I am visiting India next month and usually my PCP will asks me if I would like to get take it as a precaution for malaria - never said yes before but now wondering if I should...
They usually just give the pills for just two days I thought. Any idea on dosage and duration? I may even be able to purchase it in India

So it appears to me that Plaquenil is the same thing as the chloroquine they used in the study, which means they were testing it in combo w/ Herceptin. Excellent. Who prescribed it for you C? Your onc?

Yup, my onc. The very same forward thinker that brought me high dose Tykerb (failure) and Intrathecal Herceptin (success) - I am grateful to have such a wonderful doctor.

Sept 08-Dec 08: Herceptin +Zometa for maintenance.
Jan 09-April 09: Brain mets. Add Tykerb. Watch and wait.
April 09: Gamma Knife 10 brain mets, add Xeloda.
Sept 09: Gamma Knife to 1 brain met.
Nov 09- April 10: Lung progression, add Gemzar to Herceptin, Zometa.
May 10- Sept 10: HER2 Vaccine Trial
Sept 10: Add Tykerb for more brain mets.
Oct 10: Gamma Knife to 7 brain mets.
Dec 10: Switch from Zometa to Denosumab.
Jan 11: Gamma Knife to 3 brain mets.
March 11: Gemzar/Herceptin for lung/bone progression.
April 11: More brain mets - Intrathecal Herceptin
June 11: Ixempra/Herceptin for lung, soft tissue progression.
Aug 11: Gamma Knife
Sep 11: Abraxane/Herceptin
Future: NED

DX: 06-30-2007 - left breast -stage IIIB, Her2/Neu 3+++, ER weakly positive, PR-
Taxol+herceptin weekly for 3 months
FEC+herceptin every 3 weeks for 3 months
BRCA 1 and 2 - Negative
Jan 2008 - Bilateral mastectomy, prophylactic Rt. side.
Radiation for 5 weeks
Completed my yr of herceptin on 07-14-2008
Brain MRI - 3/2/09 Clean

There should be little or NO side effects and no problems with blood counts as a result of taking this drug, which is extracted from a bush that grows in Peru if it is the same as Plaquinil.

MY STORY SO FAR ~~~~
Found suspicious lump 9/2000
Lumpectomy, then node dissection and port placement
Stage IIB, 8 pos nodes of 18, Grade 3, ER & PR -
Adriamycin 12 weekly, taxotere 4 rounds
36 rads - very little burning
3 mos after rads liver full of tumors, Stage IV Jan 2002, one spot on sternum
Weekly Taxol, Navelbine, Herceptin for 27 rounds to NED!
2003 & 2004 no active disease - 3 weekly Herceptin + Zometa
Jan 2005 two mets to brain - Gamma Knife on Jan 18
All clear until treated cerebellum spot showing activity on Jan 2006 brain MRI & brain PET
Brain surgery on Feb 9, 2006 - no cancer, 100% radiation necrosis - tumor was still dying
Continue as NED while on Herceptin & quarterly Zometa
Fall-2006 - off Zometa - watching one small brain spot (scar?)
2007 - spot/scar in brain stable - finished anticoagulation therapy for clot along my port-a-catheter - 3 angioplasties to unblock vena cava
2008 - Brain and body still NED! Port removed and scans in Dec.
Dec 2008 - stop Herceptin - Vaccine Trial at U of W begun in Oct. of 2011
STILL NED everywhere in Feb 2014 - on wing & prayer
7/14 - Started twice yearly Zometa for my bones
Jan. 2015 checkup still shows NED
2015 Neuropathy in feet - otherwise all OK - still NED.
Same news for 2016 and all of 2017.
Nov of 2017 - had small skin cancer removed from my face. Will have Zometa end of Jan. 2018.

Good luck Courtney! I pray that this is your magic bullet that will zap those stem cells!
Thanks for unofficially trialling it for us all. Hopefully they will extend clinical trials to her 2 as well as triple negative bc soon.
Do we know if it crosses the BBB??
