Re: metformin (common anti diabetes drug) decreases her2 protein levels by 85%

Any trials available for her 2 metastatic bc?
I know Rich has long thought this could be beneficial.

Another side of the metformin discussion is that type 2 diabetes has been shown to promote HER2+ tumor development. Keeping blood sugar under good control and avoiding insulin spikes (from carb-heavy meals) is important to help avoid a recurrence. Below is a recent study on HER2 and breast cancer (Google the title if you are interested).

"Diabetes mellitus accelerates ErbB2-induced breast carcinogenesis and shortens the survival of a mouse model of breast cancer and diabetes"

Re: metformin (common anti diabetes drug) decreases her2 protein levels by 85%

If we are to take into account the conclusion of the following study metformin would be better than everolimus at inhibiting mtor.. (rapamicyn is the injection form of everolimus which is the pill form that is priced 100* more..) I wonder if then it has any effect on the immune system like everolimus/rapa?

" Although both rapamycin and metformin inhibit mTOR (the former directly and the latter through AMPK signaling), our results demonstrate previously unrecognized differences between these agents. The data are consistent with the observation that maximal induction of apoptosis and inhibition of proliferation are greater for metformin than rapamycin. "

A test to see if your tunour would respond to metformin:

Gosh I really want that test now!

Re: metformin (common anti diabetes drug) decreases her2 protein levels by 85%

Considering most therapies are a guess, might not be such a stretch to just try it without the test. Some suggest Metformin may have caloric restriction "mimetic" qualities..possibly life extending as well as weight controlling.

This appears to be the study you reference: http://www.ncbi.nlm.nih.gov/pubmed/21512313

Thanks Rich for the paper. Reading it made me think about some papers I have read recently on attacking metabolic pathways and how these can be inhibited:

A very detailed paper that describe ways to target different metabolic pathways and the various drugs that can target these (check from page 18)
