MY STORY SO FAR ~~~~
Found suspicious lump 9/2000
Lumpectomy, then node dissection and port placement
Stage IIB, 8 pos nodes of 18, Grade 3, ER & PR -
Adriamycin 12 weekly, taxotere 4 rounds
36 rads - very little burning
3 mos after rads liver full of tumors, Stage IV Jan 2002, one spot on sternum
Weekly Taxol, Navelbine, Herceptin for 27 rounds to NED!
2003 & 2004 no active disease - 3 weekly Herceptin + Zometa
Jan 2005 two mets to brain - Gamma Knife on Jan 18
All clear until treated cerebellum spot showing activity on Jan 2006 brain MRI & brain PET
Brain surgery on Feb 9, 2006 - no cancer, 100% radiation necrosis - tumor was still dying
Continue as NED while on Herceptin & quarterly Zometa
Fall-2006 - off Zometa - watching one small brain spot (scar?)
2007 - spot/scar in brain stable - finished anticoagulation therapy for clot along my port-a-catheter - 3 angioplasties to unblock vena cava
2008 - Brain and body still NED! Port removed and scans in Dec.
Dec 2008 - stop Herceptin - Vaccine Trial at U of W begun in Oct. of 2011
STILL NED everywhere in Feb 2014 - on wing & prayer
7/14 - Started twice yearly Zometa for my bones
Jan. 2015 checkup still shows NED
2015 Neuropathy in feet - otherwise all OK - still NED.
Same news for 2016 and all of 2017.
Nov of 2017 - had small skin cancer removed from my face. Will have Zometa end of Jan. 2018.

What a lively crew at SABCS!

While you are all there together, does anyone know how to make HER2 chat accessible again? It would be a great way to share our conversations on a drop-in basis live about SABCS, as well as for you all to be able to demonstrate it there at the conference as part of what HER2support can offer in connecting people who are interested in HER2 news.

Thanks for the updates. Please bring alot of transcripts back home and share them with us later. I also have been reading the Cure Magazines updates. I hope Dr. Cho, one of the oncologists I know in Hawaii brings alot of news back with him too.

1-10-2020 8mm stable nodule on R Lung, two 6mm new ones on L Lung, a possible lymph node involvement in inter fissule.

Diagnosed stage 2b in July 2003 (2.3 cm, HER2+, ER-/PR-, 7+ nodes). Treated with mastectomy (with immediate DIEP flap reconstruction), AC + T/Herceptin (off label). Cancer advanced to lung in Jan. 2007 (1 cm nodule). Started Herceptin every 3 weeks. Lung wedge resection April 2007. Cancer recurred in lung April 2008. RFA of lung in August 2008. 2nd annual brain MRI in Oct. 2008 discovered 2.6 cm cystic tumor in left frontal lobe. Craniotomy Oct. 2008 (ER-/PR-/HER2-) followed by targeted radiation (IMRT). Coughing up blood Feb. 2009. Thoractomy July 2009 to cut out fungal ball of common soil fungus (aspergillus) that grew in the RFA cavity (most likely inhaled while gardening). No cancer, only fungus. Removal of tiny melanoma from upper left arm, plus sentinel lymph node biopsy in Feb. 2016. Guardant Health liquid biopsy in Feb. 2016 showed mutations in 4 subtypes of TP53. Repeat of Guardant Health biopsy in Jana. 2021 showed 3 TP53 mutations, BRCA1 mutation and CHEK2 mutation. Invitae genetic testing showed negative for all of these. Living with MBC since 2007. Stopped Herceptin Hylecta (injection) treatment in March 2020. Recent 2021 annual CT of chest, abdomen and pelvis and annual brain MRI showed NED. Praying for NED forever!!

Background:
Despite the success of herceptin, a significant proportion of HER-2 positive breast cancer patients responded poorly to the treatment. Curcumin (diferuloylmethane), derived from Curcuma longa L,
can inhibit the formation of tumors, induce apoptosis, and act on a variety of signal transduction pathways.

Objectives:We have hypothesized that the property of curcumin to downregulate EGFR, HER-2 oncoprotein, and signal transduction pathway of PI3K/Akt, and MAPK may be used in the treatment of HER-2 over expressed breast cancer.


Results: The combination of curcumin did not interfere with the action of herceptin. The herceptin sensitive BT-474 and herceptin resistant SKBr-3(hr) were all sensitive to curcumin treatment. In western blot analysis, the phosphorylation of HER-2, Akt, MAPK were reduced in BT-474 cells when treated with herceptin. In SKBr-3(hr) cells, the addition of herceptin did not downregulate the phosphorylation of HER-2, Akt, and MAPK. When treated with curcumin, the phosphorylation of HER-2, Akt, and MAPK of SKBr-3(hr) were decreased combined with the downregulation of HER-2 oncoprotein in a dose and time dependent manner.



Conclusions: The effectiveness of curcumin in the treatment of HER-2 overexpressed breast cancer was further confirmed through in vivo BT-474 xenograft model. The comparable ability of curcumin to inhibit cell proliferation, mobility, and angiogenesis as herceptin, combined with the potential ability to overcome the herceptin resistance, curcumin may be a novel agent in the treatment of HER-2 overexressed breast cancer.

I visited that poster and brought home a reprint, but the researcher was not at that poster when I was. Would have loved to talk to them.

In case anyone does not understand what a "poster" is: At these scientific meetings studies are reduced to a large rectangular "poster" size presentation (approx 4 feet wide by 2 or 3 feet high) and put up on large cork board stands. These stands are lined up in rows with letter number IDs so we can find them from the listing in a booklet given out to attendees. There "poster sessions" in which the posters from a previous session come down and new ones go up. They try to put them in some order according to subject or type of study.
