I did some research on ONT-380. It only inhibits Her2Neu so the diarrhea is nothing like Lapatanib. It penetrates the blood brain barrier. Apparently some of the phase one studies were done on patients with brain mets. Ont=380 was given combined with other drugs. None of the patients with brain mets progressed. There was one remission, one partial remission and all of the other patients with brain mets stayed stable.

The dosage given was 300mg orally twice daily. The side effects were nausea, some diarrhea rash and increase in liver enzymes.
